Introduction:
NSI-189 is an intriguing neurotrophic compound which has shown potential to upregulate neural growth factors (most notably BDNF and SCF, among others), stimulating the creation of new neurons and synapses, particularly within the hippocampus. This drug, however, is still relatively new and lacks much anecdotal or medical literature, making it a risky choice until more trials are undergone and its true practicality is determined. In this thread, I'll discuss its supposed mechanism of action and potential use cases.

How It Works:
This compound works by raising available levels of the aforementioned growth factors (BDNF, GDNF, SCF, etc), meaning higher levels of these factors are secreted by various cells within the brain. The elevated levels of these growth factors allow for more agonization of the appropriate receptors (cba to list them) and indirectly cause an increase in neuron and neural pathway proliferation. Aptly; the drug increases endogenous secretion of neural growth factors, indirectly increasing neuro & synaptogenesis. This means it may positively impact your cognition long term as a function of more neurons and synapses within the brain.

The reason for the compound's selectivity towards the hippocampus in practice is largely because of its relatively high concentration of STEM (unspecialized) cells, which allows for heightened neurogenesis specifically within this region of the brain. This is a common theme amongst neurotropic compounds (ie. cerebrolysin). The selectivity towards the hippocampus by NSI-189 and other neurotrophic agents implies that its effects on cognition would primarily impact memorization, as well as spatial navigation and emotional regulation, to a lesser degree. (study 1 , study 2)

Why It Might Be Useful:
Overall, this molecule may be useful for anyone who wants to improve their cognition by way of memorization skills or emotional regulation. Additionally, NSI has been seen to show some efficacy in treating patients with MDD (major depressive disorder, see study 2). The oral route seen in the current literature may also make it more convenient than similar compounds which must be administered via injection. Due to its neruo and synaptogenerative effects, it could also have the potential to be used for neuroprotection, cognitive recovery, or exploited to increase neuroplasticity.

Closing Statements:
I hope anyone who read to the end enjoyed and maybe learned something new. This is my first informational thread, so any constructive criticism would be appreciated. Everything is based on publicly available studies (via pubmed), however my explanation is quite oversimplified so do your own research. Also, please consider the fact that this is still an experimental compound; again, do your own research! Lastly, shout out Biomaxx Biomaxx for putting me onto this compound.

mirin, please make font size just a bit bigger next time:pepecheers:
although its difficult to source, how well do most nootropics including this compare to crebinostat?
and for that matter, how well do most nootropics compare to microdosed psychedelics?

Honestly I couldn't say since I haven't given it much research, but I can look into creb and give you my opinion sometime soon. I've heard good things about psychs like psylocibin being efficacious for increasing neural plasticity in therapeutic dosages, but my personal experience can't exactly speak to that 😭

i appreciate you putting in the work. ive never really looked into nootropics since i havent really had any academic struggles except having shit concentration and some other stuff in class:feelsokman:

what nootropics have you personally used?
