What It Is

Mifepristone (RU-486), is a glucocorticoid receptor antagonist. It doesn’t lower cortisol production, it blocks cortisol from actually binding to its receptors in tissues, including bone.

How it works

Cortisol is highly catabolic to bone. It inhibits osteoblasts (the cells that build bone) and promotes bone breakdown. Mifepristone binds to the glucocorticoid receptor and prevents cortisol from doing its damage. This creates a more anabolic environment for bone tissue without directly raising GH or testosterone.

Why it matters for looksmaxxing

• Reduces cortisol.

• May support better bone density and periosteal growth (thicker bones, wider frame).

• Mifepristone directly blocks the glucocorticoid receptor (actual cortisol signaling).
This makes it more targeted at protecting bone tissue.

Doses

25–50 mg per day, taken in the morning.

Most start at 25 mg and only go to 50 mg if tolerated well.

Cycles are usually kept short: 6–12 weeks on, then equal time off.

Taken with food to reduce stomach discomfort.

Side Effects

• Can lower testosterone due to mild anti-androgen activity.

• May shift hormonal balance slightly toward estrogen, Able to counter it with an AI.

• Possible fatigue or mood changes.

• Not a lot of research or talked about.

Countering Risks

• Use the lowest effective dose (start at 25 mg).

• Pair it with enclomiphene if running any androgens.

• Do not run it year-round.

Conclusion

Mifepristone is one of the most niche pharma for someone trying to protect, build bone mass, reduce cortisol during their growth years. It doesn’t raise GH or testosterone, but it removes one of the biggest brakes on bone formation that is cortisol signaling. While not a mainstream looksmax compound and definitely not risk free, it remains one of the few orals that directly targets cortisol at the receptor level.

WOW a Peptide that gets rid of cortisol 🗿🗿🗿🍷🍷🍷🍷
small molecule actually
Relacorilant is more selective when blocking cortisol. very weak on anti-progesterone effect and is really minimal on anti androgen effects and has much less shift in estrogen shift so it’s better. but relacorilant is much newer with less research
yea relacorilant mogs then
literally no one cares about how researched something is
