Sustained radial dilation activates piezo1/2 ion channels in sphincter sarcolemma.
This triggers Ca²⁺ influx → calcium/calmodulin-dependent protein kinase II (CaMKII) phosphorylation → nuclear translocation of YAP/TAZ (Hippo pathway off-switch).
YAP/TAZ upregulates RUNX2 and SP7 (Osterix) in mesenchymal stem cells (MSCs) resident in pelvic periosteum.
