In my attempt of a BOTB thread I will dive into fgfr3 inhibitors.
This thread will cover:
-What they are
-How they can help
-Studies
-Side effects
fgfr3 inhibitors are usually oral tablets designed to block fibroblast growth factor receptor 3 signalling (FGFR3). Fgfr 3 common FDA approved drugs are erdafitnib and pemigatinib. Pemigatinib is commonly used for treating bile duct cancer and erdafitinib is commonly used for treating cancer lining the bladder.
The overall response rate for fgfr 3 is approximately 20 - 40%.
Another purpose of fgrfr 3 inhibitors is to regulate bone growth which is where they can help.
FGFR 3 acts like a brake when growing bones however it is not negative. When FGFR 3 is strong bone growth slows, when its blocked growth plates can produce more cartilage therefore creating longer bones.
Therefore in theory FGFR 3:
- Increase bone growth
-Slightly affect limb proportions and help frame
-Influence facial skull growth during development
-Help with height
However, this works best while growth plates are open. When they close (16 - 18 females, & 18-21 males) growth from these drugs become impossible. Neither will it help sculpt the skull in adults only teenagers.
Studies show that with daily injections before growth plates close height can grow between 1.5 - 2 cm every year.
Studies also show:
- Forward growth of the midface
- Better orbital positioning
-Better chin projection
These changes are small yet noticeable
Although we may not care here are some side effects which can be experienced while taking fgfr3 inhibitors
Injection site reactions such as swelling, redness, itching with an approx. 85% chance of happening
Immunity to the drug with 35% chance
vomiting with 27% cahnce
joint pain with 15% chance
low blood pressure with 13% chance
